Police said the driver, Kabir O'Farril Diaz, 38, and passenger, Terry Lynn Kimball, 56, started arguing about the route Diaz was taking.
Diaz pulled over at a convenience store on 58th Street North and the men had a "physical conversation," police said.
According to spokeswoman Yolanda Fernandez, witnesses told officers the driver wasn't the aggressor during the fight that involved punching and kicking.
Detectives are reviewing surveillance video in the area to verify if that is the case.
Fernandez said investigators are looking into whether the driver's only punch was done in self-defense.
Fernandez added there was a language barrier between the two men, as the Uber driver only spoke Spanish.
Detectives are also waiting on the toxicology test results on the passenger before finishing their investigation.
Officials said Diaz has cooperated with the investigation and no charges have been filed.
